RESUMO

Abstract The present study involves the chemical and bacteriological analysis of water from different sources i.e., bore, wells, bottle, and tap, from Peshawar, Mardan, Swat and Kohat districts of Khyber Pakhtunkhwa (KP) province, Pakistan. From each district, 50 water samples (10 samples from each source), regardless of urban and rural status, were collected from these sources and analysed for sulphates, nitrates, nitrites, chlorides, total soluble solids and coliforms (E. coli). Results indicated that majority of the water sources had unacceptable E. coli count i.e.> 34 CFU/100mL. E. coli positive samples were high in Mardan District, followed by Kohat, Swat and Peshawar district. Besides this, the some water sources were also chemically contaminated by different inorganic fertilizers (nitrates/nitrites of sodium, potassium) but under safe levels whereas agricultural and industrial wastes (chloride and sulphate compounds) were in unsafe range. Among all districts, the water quality was found comparatively more deteriorated in Kohat and Mardan districts than Peshawar and Swat districts. Such chemically and bacteriologically unfit water sources for drinking and can cause human health problems.

Chromium (VI) a highly toxic metal, a major constituent of industrial waste. It is continuously release in soil and water, causes environmental and health related issues, which is increasing public concern in developing countries like Pakistan. The basic aim of this study was isolation and screening of chromium resistant bacteria from industrial waste collected from Korangi and Lyari, Karachi (24˚52ʹ46.0ʺN 66˚59ʹ25.7ʺE and 24˚48ʹ37.5ʺN 67˚06ʹ52.6ʺE). Among total of 53 isolated strains, seven bacterial strains were selected through selective enrichment and identified on the basis of morphological and biochemical characteristics. These strains were designated as S11, S13, S17, S18, S30, S35 and S48, resistance was determined against varying concentrations of chromium (100-1500 mg/l). Two bacterial strains S35 and S48 showed maximum resistance to chromium (1600 mg/l). Bacterial strains S35 and S48 were identified through 16S rRNA sequence and showed 99% similarity to Bacillus paranthracis and Bacillus paramycoides. Furthermore, growth condition including temperature and pH were optimized for both bacterial strains, showed maximum growth at temperature 30ºC and at optimum pH 7.5 and 6.5 respectively. It is concluded that indigenous bacterial strains isolated from metal contaminated industrial effluent use their innate ability to transform toxic heavy metals to less or nontoxic form and can offer an effective tool for monitoring heavy metal contamination in the environment.

Abstract Background Ectodermal dysplasia syndactyly syndrome 1 (EDSS1) is a rare hereditary disorder characterized by defects in teeth, hair, and nails in association with a fusion of the digits. Genetically, the disease phenotypes are caused by homozygous and compound heterozygous variants in NECTIN4 gene. Objective The main objective of the study was to identify the pathogenic sequence variant(s) for family screening and identification of carriers. Methods In the present study, the authors have investigated a large consanguineous family of Pakistani origin segregating autosomal recessive EDSS1. All the coding exons of the NECTIN4 gene were directly sequenced using gene-specific primers. Results The affected individuals presented the classical EDSS1 clinical features including sparse hair, hypoplastic nails with thick flat discolored nail plates, peg-shaped, conical, and widely spaced teeth with enamel hypoplasia, proximal cutaneous syndactyly of fingers and toes. Sequence analysis of the coding region of the NECTIN4 identified a novel nonsense variant [c.163C>T; p.(Arg55*)] in exon-2 of the gene. Computational analysis of protein structure revealed that the variant induced premature termination at Arg55 located in Ig-like V-loop region leading to loss of Ig-C2 type domains and transmembrane region, and most likely Nectin-4 function will be lost. Study limitation Gene expression studies are absent that would have strengthened the findings of computational analysis. Conclusion The present study expanded the phenotypic and mutation spectrum of the NECTIN4 gene. Further, the study would assist in carrier testing and prenatal diagnosis of the affected families.

RESUMO

Many pathogenic strains have acquired multidrug-resistant patterns in recent a year, which poses a major public health concern. The growing need for effective antimicrobial agents as novel therapies against multidrug-resistant pathogens has drawn scientist attention toward nanotechnology. Silver nanoparticles are considered capable of killing multidrug-resistant isolates due to their oligo-dynamic effect on microorganisms. In this research study NPs were synthesized using the gram-positive bacteria Lactobacillus bulgaricus and its activity against selected pathogenic strains. Lactobacillus bulgaricus pure cultures were isolated from raw milk and grown in "De Man, Rogasa, and Sharp" broth for synthesis of nanoparticles. Lactobacillus bulgaricus culture was centrifuged and Cell free supernatant of it was employed with aqueous silvery ions and evaluated their antibacterial activities against bacterial strains i.e. Staphylococcus aureus, Staphylococcus epidermidis and Salmonella typhi using agar well diffusion assay. Antibiotic profiling against selected pathogenic strains were also conducted using disc diffusion method. The synthesis and characterization of silver nanoparticles were monitored primarily by the conversion of the pale-yellow color of the mixture into a dark-brown color and via ultraviolet-visible absorption spectroscopy and Scanning electron microscopy respectively. The result showed that that AgNPs with size (30.65-100 nm) obtained from Lactobacillus bulgaricus were found to exhibit antibacterial activities against selected bacterial strains. Taken together, these findings suggest that Lactobacillus bulgaricus has great potential for the production of AgNPs with antibacterial activities and highly effective in comparison to tested antibiotics.

The emergence of multi-drug resistant (MDR) bacterial strains, which are posing a global health threat has developed the interest of scientists to use bacteriophages instead of conventional antibiotics therapy. In light of an increased interest in the use of phage as a bacterial control agent, the study aimed to isolate and characterize lytic phages from sewage effluent. During the current study, bacteriophage AS1 was isolated from sewage effluent against E.coli S2. The lytic activity of phageAS1 was limited to E.coli S2 strain showing monovalent behavior. The calculated phage titer was 3.5×109 pfu/ml. PhageAS1 was stable at a wide range of pH and temperature. The maximum stability was recorded at 37ºC and pH 7.0, while showing its normal lytic activity at temperature 60ºC and from pH 5.0 to 11.0 respectively. At temperature 70ºC, phage activity was somewhat reduced whereas, further increase in temperature and decrease or increase in pH completely inactivated the phage. From the current study, it was concluded that waste water is a best source for finding bacteriophages against multi-drug resistant bacterial strains and can be used as bacterial control agent.

Young males and females are highly concerned about their body image and physical appearance. They refrain themselves from eating that effect the physical and social well-being. This study aims at identifying the presence of eating disorder and to explore as well as confirm four factor solution of eating disorder among adolescents and adults. The study is conducted obtaining data from a sample of 600 adolescents and young adults studying in private and public educational institutions using EDE-Q questionnaire. Chi-square test has revealed the relationship between age and eating concern scores. Pearson correlation test suggested that age is related with global score whereas all the eating disorder scores are inter-related. No gender-wise difference between shape concern and global score is found whereas the global scores and shape concern scores are significantly different with respect to type of institution. Global score and shape concern scores are same for adolescents and young adults. Mean ranks are higher for males compared to females whereas there is significant difference among restraint scores gender-wise however shape concern, weight concern and restraint scores were significantly different in private and public institutions. Mean ranks for young adults are higher for restraint, shape concern, weight concern and eating concern scores compared to adolescents whereas the significant difference was observed between young adults and adolescents for restraint scores. EFA has identified four-factor solution for the EDE-Q subscales whereas CFA has also confirmed it. The four factor solution is weight and shape concern, fear of gaining weight, restraint and eating concern.

RESUMO

Background: The thyroid gland is endocrine gland located in front and lower side of neck. Thyroid gland secretes two types of thyroid hormones that are triiodothyronine (T3) and tetraiodothyronine (T4). The hypothalamus is a center for regulation of thyroid hormones. It senses the low hormone levels and in turn releases thyrotropin-releasing hormone (TRH). TRH stimulates anterior pituitary to release TSH which then acts on the thyroid gland to maintain normal level of T3 and T4. The objective of study is to determine the effects of carbamazepine on TRH in euthyroid rabbits. Methods: An experimental study performed on 30 rabbits. These were divided into three groups having 10 rabbits in each group. 10 rabbits were treated with 10mg/kg/day of CBZ (OD), other 10 with 35mg/kg/day CBZ (three divided doses) and 10 rabbits served as control. T3, FT4, TSH and TRH levels were evaluated at baseline and after 21 days of treatment in all three groups by Electro-chemiluminescence immunoassay and ELISA respectively. Results: Comparison of the hormone levels of the control group and the group having a dose of 10 mg/kg/day for 21 day of treatment. Comparative results showed serum level of T3 (P=0.031), FT4 (P=0.030), and TRH (P=0.044) levels significantly lower than the control group and TSH (P=0.057) levels remain unaltered. It was also found that group having a dose of 35 mg/kg/day; TDS showed decrease in T3 (P value 0.001), FT4 (P=0.001), TSH (P=0.003) and TRH (P=0.001) level as compared to control group. Conclusions: Our data suggest that Carbamazepine monotherapy does alter thyroid hormones and its central regulatory hormone TRH. Decrease in TRH level increase level of depression and suicidal thoughts and also risk of tertiary hypothyroidism. These findings could have very important clinical implications.

RESUMO

Dyke-Davidoff-Masson Syndrome [DDMS] is a rare neurological condition characterised by drug-resistance seizures, hemiparesis, mental retardation, facial asymmetry, and intellectual disabilities. On brain imaging, the disease is characterised by cerebral hemiatrophy with ipsilateral calvarial thickening and hyperpneumotisation of paranasal sinuses or mastoid air cells. Although more common in men and on the left side of the brain, the disease can affect both genders and cerebral hemispheres. It mainly presents in childhood. The adult presentation is unusual but has been reported in medical literature. Most of the patients need more than one antiepileptic agent for optimal control of seizures. Hemispherectomy is reserved for patients who have drug-resistant and disabling seizures. The good prognostic factors are disease onset before age of two and better seizure control. We report two cases of DDMS occurring in teenage boys who presented with status epilepticus. The clinical histories, radiological findings, and treatment of both patients are discussed below

RESUMO

The 1,2,3-triazole-containing [1-azido-1-deoxy-beta -D-glucopyranoside] complex was synthesized using click chemistry approach and evaluated its potential as a tumor-seeking agent. In the present study, [99m] Tc-tricarbonyl labeled [1-azido-1-deoxy-beta -D-glucopyranoside] radiotracer [[99m] Tc[CO][3]-BM], [where BM stands for biomolecule, e.g., [1- azido-1-deoxy-beta -D- glucopyranoside]] was synthesized via click chemistry approach and then labeled with technetium- 99m through isolink kit. Radio labeled drug was tested for radiochemical purity and in vitro stability by chromatographic techniques. Normal distribution and tumoral uptake were studied in Swiss Webster mice. Radiochemical purity results show 97.9 +/- 1.5% labeling and its in vitro stability were studied at room temperature up to 5h. The radio labeled drug exhibited 73.6 +/- 1.1% binding with blood proteins. Normal distribution of drug shows prominent uptake in brain while in case of tumor-bearing mice, the uptake was maximum in tumor tissue and negligible amount was shown in brain. The biodistribution was further compared with 2-fluoro-2-deoxy glucose [[18]F-FDG], which was used as a positive control. The data indicate that [99m] Tc-tricarbonyl labeled [1-azido-1-deoxy-beta -D- glucopyranoside] radiotracer might be a feasible candidate with reasonable potential for tumor diagnosis

RESUMO

Textiles perform as an interface between the body and environment. In the new generation of textiles, this potential of textiles is being utilized to provide the resistance not only against weather but also against micro-organisms and collision forces etc. the type of weave has a strong impact on its physical properties. Type of weave also affects the suitability of the material end use. According to a study by Behera plain weave is more suitable for surgical apparels in comparison with twill weave because of large pores between two adjoining yarns on intersection. Fabric construction parameters determine the strength of the fabric. Parameters are: mass per unit area, number of threads per unit length, maximum force using the strip method, and yarn count [yarn number]. A fabric with high thread count does not allow the microorganisms to pass through a fabric than a fabric having low/less count. Different Surface finishes help to decrease the bacterial growth on the surface of fabric. Plasma coated Non-woven samples are better repellent of pathogens than re-usable woven fabrics. All surgical attire has to meet some given standards for tensile strength which dictates the rate of wear and tear that will affect the required properties. The thought behind this study was to evaluate and compare the tensile strength of existing surgical gowns among the groups. 1] To find out the Tensile Strength of existing surgical gown fabric before and after laundering [among three groups of hospitals]. 2] To compare the results of Tensile Strength of existing surgical gowns [among three groups of hospitals] being used in hospitals of Lahore. The study has experimental design. March 2011- March. 2012. Samples of fabric used for making surgical gowns were collected from different hospitals of Lahore and evaluated for Tensile Strength before and after laundering at Textile Lab. On the whole a decreasing trend was observed for tensile strength in all 3 groups of hospitals from 0 to 20[th] laundering. Group B of hospitals showed lowest tensile strength observation whereas the highest value was observed in group A of hospitals. The lowest observation for tensile strength was found in Group B of hospitals whereas in group C the highest value was observed it's clear that fabrics of group B showed lowest tensile strength rating in test results for both warp and weft directions. Whereas group A of hospitals showed highest tensile strength test results among the groups in warp and in weft group C showed high results
